Chemical protein synthesis may provide proteins that cannot be obtained recombinantly. Although a number of proteins have been synthesized through total synthesis, there are still many proteins that we just cannot make. In many occasions we even cannot either predict where or understand why the synthesis may fail. In the meeting I wish to present some of our studies on the chemical synthesis of difficult proteins, including membrane proteins, proteins with lipid modifications such as pamitolytion, and proteins over 300 amino acids. I will discuss where we observe troubles and how we have tried to overcome some of the problems.
